چکیده

Adenylate cyclase has been found to be associated with par t i cu la te fractions from various m a m m a lian cells and unicellar organisms (for review see reference 16). Mostly, the enzyme has been associated with the plasma membrane , a l though in brain it was conta ined in the synaptic membranes (3), in the rabbi t psoas muscle in the microsomal fraction (15), and in the lymphocyte on the plasma and nuclear membranes (17). Al though hypotha lamic releasing factors seem to st imulate the secretion of certain hormones from adenohypophysis through adenylate cyclase and cyclic A M P (16), the subcellular dis tr ibut ion of the enzyme has not been studied. This prel iminary communica t ion reports the finding of marked enr ichment of adenylate cyclase in the p lasma membrane and Golgi fract ions of porcine adenohypophysis . These subcellular fract ions were each enriched in their respective components but cross-contaminat ion did exist as shown by electron microscope and enzyme marker studies. Synthesis of Plasmalemmal Glycoproteins in Intestinal Epithelial Cells

The relationship between Golgi and cell surface membranes of intestinal cells was studied. These membranes were isolated from intestinal crypt cells and villus cells. The villus cell membranes consisted of microvillus membrane, a Golgi-rich fraction, and two membrane fractions interpreted as representing lateral-basal membranes. The villus cell microvillus membrane was purified by previously pu...

Purification of adenohypophyseal plasma membranes and properties of associated adenylate cyclase.

Plasma membranes were purified from bovine anterior pituitary tissue by differential and isopycnic centrifugation according to a modification of Neville’s technique (Biophys. Biochem. Cytol. 8, 413, 1960). Plasma membranes, rough endoplasmic reticulum, and secretory granules sedimented at the interfaces of sucrose layers of different densities and were thus simultaneously obtained in high yield...

Presence of adenylate cyclase activity in Golgi and other fractions from rat liver. I. Biochemical determination

The distribution of adenylate cyclase (AC) in Golgi and other cell fractions from rat liver was studied using the Golgi isolation procedure of Ehrenreich et al. In liver homogenate the AC activity was found to decay with time, but addition of 1 mM EGTA reduced the rate of enzyme loss. The incorporation of 1 mM EGTA into the sucrose medium used in the initial two centrifugal steps of the Golgi i...

Activation of spermatozoan adenylate cyclase by a low molecular weight factor in porcine seminal plasma.

The ejaculated porcine spermatozoa were fractionated into the cytosol, membrane, midpiece plus tail (flagella) and head fractions, and their adenylate cyclase activities were measured. About 65% of the total activity was located in the flagella fraction. For all the fractions, Mn2+-dependent adenylate cyclase activity was about 20 times higher than Mg2+-dependent activity, and guanine nucleotid...

Presence of adenylate cyclase activity in Golgi and other fractions from rat liver. II. Cytochemical localization within Golgi and ER membranes

The presence of adenylate cyclase (AC) in liver Golgi and microsomal fractions from ethanol-treated rats was tested cytochemically using 5'-adenylyl imidodiphosphate (AMP-PNP) lead phosphate method. Parallel biochemical assays showed that rat liver Golgi AC was only partially inhibited by lead: in the presence of 1 mM Pb++ 80% of the enzyme was preserved, while when 2 mM Pb++ was used 25% remai...
